细胞抗氧化英语《Cellular Antioxidants: The Key to Fighting Oxidative Stress》Oxidative stress is a process that occurs in the body when there is an imbalance between free radicals and antioxidants. Free radicals, which are highly reactive molecules, can cause damage to cells and tissues, leading to various health issues such as inflammation, aging, and chronic diseases. Antioxidants play a crucial role in combating oxidative stress by neutralizing free radicals and protecting the body from their harmful effects.Within the body, cells are constantly exposed to oxidative stress due to various factors such as pollution, UV radiation, and poor diet. As a result, cells rely on antioxidants to protect themselves and maintain proper functioning. Cellular antioxidants, which include enzymes like superoxide dismutase and catalase, as well as nutrients like vitamins C and E, are essential in preventing cellular damage caused by free radicals. One of the key functions of cellular antioxidants is to scavenge free radicals and prevent them from causing oxidative damage. When cells are exposed to oxidative stress, antioxidants work to neutralize free radicals and reduce their harmful effects. This not only protects the cells from damage but also helps to maintain proper cellular function and overall health. In addition to their scavenging properties, cellular antioxidants also play a role in repairing oxidative damage. When cells are damaged by free radicals, antioxidants can help to repair the damage and restore the cell to its normal state. This process is crucial for maintaining the integrity of cellular structures and preventing further damage.Furthermore, cellular antioxidants are involved in regulating the body's response to oxidative stress. This includes activating various cellular defense mechanisms and signaling pathways that help to mitigate the effects of oxidative stress. By doing so, antioxidants help to protect cells from oxidative damage and maintain their overall integrity. In conclusion, cellular antioxidants play a crucial role in fighting oxidative stress and protecting the body from the harmful effects of free radicals. By scavenging free radicals, repairing oxidative damage, and regulating cellular responses to oxidative stress, antioxidants are essential for maintaining cellular function and overall health. Therefore, it is important to consume a diet rich in antioxidants and to engage in healthy lifestyle practices that can help to support the body's natural antioxidant defenses.。

Ethnobotanical Leaflets 12: 1172-75. 2008.Anti-hyperglycaemic and Insulin Release Effects of Coccinia grandis (L.) VoigtLeaves in Normal and Alloxan Diabetic RatsA. Doss and R. DhanabalanPG Department of MicrobiologyRVS College of Arts and Science, Coimbatore, Tamil Nadu, IndiaCorrespondingauthor:*******************Issued 15 December 2008ABSTRACTCoccinia grandis (L.) Voigt (Cucurbitaceae), occurs throughout the world and has intensive popular use in the treatment of infections. The main aim of the present work was to investigate the antidiabetic effects of aqueous extracts of leaves of C. grandis obtained by Decoction method. Graded doses of the aqueous extract were administered to normal and experimental diabetic rats for 10 days. Significant (p < 0.05) reduction in fasting blood glucose levels were observed in the normal as well as in the treated diabetic animals. Serum insulin levels were not stimulated in the animals treated with the extract. The changes in body weight, serum lipid profiles, liver glycogen levels were assessed in the extract treated diabetic rats and compared with diabetic control and normal animals.Key words: Antidiabetic activity, alloxan monohydrate, Gliben clamide, Coccinia grandis.INTRODUCTIONDiabetes mellitus (DM) is a serious health problem with high rates of incidence and mortality. DM is characterized by elevated plasma glucose concentrations resulting from insufficient insulin, insulin resistance or both leading to metabolic abnormalities in carbohydrates, lipids and proteins (Hernandez-galicia et al., 2002). According to the World Health Organization, more than 70% of the world’s population must use traditional medicine to satisfy their principal health needs (Farnsworth et al., 1985). A great number of medicinal plants used in the control of the DM have been reported (Baily & Day 1989; Marles & Farnsworth, 1994). However these plants represent alternatives to developing new oral hypoglycemic agents, appropriate ethnobotanical information is scarce, obscure and ambiguous.Coccinia grandis (L.) Voigt. (Family: Cucurbitaceae) is a climbing perennial herb distributed almost all over the world. The leaves of the plant possess antidiabetic, anti-inflammatory, antipyretic, analgesic, antispasmodic, antimicrobial, cathartic and expectorant activities (Asolkar et al, 1992; Nadkami and Nadkami, 1992).The leaves contain Triterpenoids, alkaloids and tannins (Rastogi. and Mehrota, 1990).The objective of the present work is to make an analysis of the ethnobotanical information on Coccinia grandis used in South Indian region for diabetes mellitus control.MATERIAL AND METHODSPlant material and decoction preparationThe leaves of C. grandis were collected in December 2007 from Coimbatore district, Tamilnadu, India and authenticated by Botanical Survey of India (Southern circle), Coimbatore, Tamil Nadu, India. A voucher specimen is being maintained in the RVS College of Arts and Science and RVS Medical & Ayurvedic foundation, Coimbatore. The leaves were shade dried and powdered.About 150 g of dried powdered leaves were boiled in 1 liter of water for 5 min, allowing the decoction to stand for 30 min and filtered through Whatman no.1 filter paper which yielded a decoction with a 15% higher concentration than that of produced by the method described by Teixeira et al., (1990).Test animalsThe test animals used in the study were procured from Karpagam Medical and Research Foundation. Six Albino rats of wistar strain weighing 150 - 200 g bred in Animal Tissue Culture Lab, Karpagam Arts and Science College. They were individually housed in polypropylene cages in well-ventilated rooms, under hygienic conditions. Animals were given water ad libitum and fed with rat pellet feed.Induction of Experimental Diabetes and TreatmentAlloxan monohydrate solution of 10 mg/ml was prepared in ice-cold citrate buffer 0.1 M, pH 4.5 and was administered to the rats within 5 min at a dose of 50 mg/kg body weight intraperitonially. After 48 h of alloxan monohydrate administration, rats with moderate diabetes having glycosuria and hyperglycemia were taken for the experiment.Rats weighing 150 – 160 g, fasted over night were used for induction of diabetes. Rats were divided into two sets; diabetic and non-diabetic. Group I received normal diet and served as normal control. Group II consists of alloxan-induced rats receiving normal diet and serving as diabetic control. Group III consists of alloxan induced rats receiving Gliben clamide (synthetic antidiabetic drug) at 0.5 mg/kg body weight once a day orally for 10 days. Group IV consists of alloxan-induced rats receiving C. grandis (1 ml) once a day orally for 10 days. Group V consists of normal rats receiving C. grandis (1 ml) once a day orally for 10 days.Blood samples were collected through the tail vein just prior to and on days 10 after drug administration. The blood glucose, urea, cholesterol, serum glutamate oxygenate transaminase (SGOT) and serum glutamate pyruvate transaminase (SGPT) were determined for all the samples.StatisticsThe data were analyzed using one-way ANOVA followed by Dunnett's test. The level of significance was set at 0.05.RESULTS AND DISCUSSIONThe extracts of C. grandis produced significant changes in the alloxan-induced diabetic rats (Table 1). The aqueous extracts of C. grandis reduced the glucose levels considerably. Treatment of the diabetic rats with Gliben clamide (10 mg/kg) also reduced blood glucose level. The prolonged treatment of C. grandis extracts on alloxan-induced diabetes rats produced consistent reduction in the blood glucose levels. The aqueous extract also reduced urea, protein and cholesterol during the 10 days treatment period (Tables 1 and 2).It is possible that the drug may be acting by potentiating the pancreatic secreation or increasing the glucose uptake. Hypercholesterolemia, hypertriglyceridemia and hyperurea have been reported to occur in alloxan diabetic rats (Resmi et al., 2001; Joy and Kuttan, 1999). Increase in glycogen in liver can be brought about by an increase in glycogenesis and/or decrease a glycogenolysis. Therefore, the C. grandis extract could have stimulated glycogenesisand/or inhibited glycogenolysis in diabetic rat liver. The plant extracts treated animals showed non-toxicity of the extract, which indicates that unlike insulin and other common hypoglycemic agents, overdose of the drug may not result in hypoglycemia. The increase in total protein (Table 2) may be due to changes in circulating amino acids levels, hepatic amino acids uptake, and muscle output of amino acid concentrations (Felig et al., 1977). The non-protein nitrogen compound urea is found to be increased when compared to plant extract treated rats. The level of SGPT and SGOT increased remarkably in the C. grandis extract-treated rats (Nagappa et al., 2003). Our results support Ghosh et al., (2004) who reported that transaminase activity is increased in serum of diabetics. In diabetic animals, the changes in level of serum enzymes are directly related to changes in the metabolism (Felig et al., 1977).C. grandis leaf extract showed significant anti-diabetic effect in diabetic rats after oral administration. Thus, the claim made by the Indian systems of medicine regarding the use of leaf extract of this plant in the treatment of diabetes is validated. Present efforts are directed to isolate the active constituents and elucidation of mechanism of action.ACKNOWLEDGEMENTSThe authors express their sincere gratitude to the management of RVS College of Arts and Science, Coimbatore.REFERENCE1. 英语Antioxidant Mechanisms of Action.Antioxidants are molecules that prevent or slow down oxidation, a chemical reaction that produces free radicals and can damage cells. Free radicals are unstable molecules with unpaired electrons that can react with other molecules and cause damage to the cell's structure and function.Antioxidants work by neutralizing free radicals and preventing them from causing damage. They do this by donating an electron to the free radical, which stabilizes the free radical and prevents it from reacting with other molecules. There are many different types of antioxidants, each with its own unique mechanism of action. Some of the most common types of antioxidants include:Vitamin E: Vitamin E is a fat-soluble antioxidant thatprotects cell membranes from damage by free radicals. It works by donating an electron to the free radical, which stabilizes the free radical and prevents it from reacting with the cell membrane.Vitamin C: Vitamin C is a water-soluble antioxidant that protects cells from damage by free radicals. It works by donating an electron to the free radical, which stabilizes the free radical and prevents it from reacting with other molecules.Beta-carotene: Beta-carotene is a fat-soluble antioxidant that protects cells from damage by free radicals. It works by donating an electron to the free radical, which stabilizes the free radical and prevents it from reacting with other molecules.Polyphenols: Polyphenols are a group of antioxidants that are found in many fruits and vegetables. They work by donating an electron to the free radical, which stabilizes the free radical and prevents it from reacting with other molecules.Antioxidants are important for protecting cells from damage by free radicals. They can help to prevent or slow down the development of many chronic diseases, such as cancer, heart disease, and Alzheimer's disease. How to Get Antioxidants.Antioxidants can be obtained from a variety of foods, including:Fruits and vegetables.Whole grains.Nuts and seeds.Beans and legumes.Tea and coffee. It is important to eat a variety of foods to get a widerange of antioxidants. You can also take antioxidant supplements, but it is important to talk to your doctor before starting any supplements.Benefits of Antioxidants.Antioxidants have many health benefits, including:Reducing the risk of chronic diseases, such as cancer, heart disease, and Alzheimer's disease.Protecting cells from damage by free radicals.Improving immune function.Slowing down the aging process.Improving skin health.Risks of Antioxidants.Antioxidants are generally safe, but there are somepotential risks, including:Some antioxidants can interact with medications, so it is important to talk to your doctor before starting any supplements.High doses of antioxidants can actually be harmful, so it is important to take them in moderation.Conclusion.Antioxidants are important for protecting cells from damage by free radicals. They can help to prevent or slow down the development of many chronic diseases. Antioxidants can be obtained from a variety of foods, including fruits, vegetables, whole grains, nuts, seeds, beans, legumes, tea, and coffee.

Antiapoptotic and Antiautophagic Effects of Glial Cell Line-Derived Neurotrophic Factor and Hepatocyte Growth Factor After Transient Middle Cerebral Artery Occlusion in RatsJingwei Shang,1Kentaro Deguchi,1Toru Yamashita,1Yasuyuki Ohta,1Hanzhe Zhang,1Nobutoshi Morimoto,1Ning Liu,1Xuemei Zhang,1Fengfeng Tian,1Tohru Matsuura,1Hiroshi Funakoshi,2Toshikazu Nakamura,3and Koji Abe1*1Department of Neurology,Okayama University Graduate School of Medicine,Dentistry and Pharmaceutical Sciences,Okayama,Japan2Division of Molecular Regenerative Medicine,Department of Biochemistry and Molecular Biology, Osaka University Graduate School of Medicine,Osaka,Japan3Kringle Pharma Joint Research Division for Regenerative Drug Discovery,Center for Advanced Medicine,Osaka University,Osaka,JapanGlial cell line-derived neurotrophic factor(GDNF)and he-patocyte growth factor(HGF)are strong neurotrophic factors, which function as antiapoptotic factors.How-ever,the neuroprotective effect of GDNF and HGF in ameliorating ischemic brain injury via an antiautophagic effect has not been examined. Therefore,we investi-gated GDNF and HGF for changes of infarct size and antiapoptotic and antiautophagic effects after transient middle cerebral artery occlusion(tMCAO)in rats. For the estimation of ischemic brain injury, the infarct size was calculated at24hr after tMCAO by HE staining.Terminal deoxynucleotidyl transferase-mediated dUTP-biotin in situ nick end labeling(TUNEL)was performed for evalu-ating the antiapoptotic effect.Western blot analysis of microtubule-associated protein1light chain3(LC3)and immunofluorescence analysis of LC3and phosphoryl-ated mTOR/Ser2448(p-mTOR)were performed for evalu-ating the antiautophagic effect.GDNF and HGF signifi-cantly reduced infarct size after cerebral ischemia. The amounts of LC3-I plus LC3-II(relative to b-tubulin)were significantly increased after tMCAO,and GDNF and HGF significantly decreased them.GDNF and HGF sig-nificantly increased p-mTOR-positive cells.GDNF and HGF significantly decreased the numbers of TUNEL-, LC3-,and LC3/TUNEL double-positive cells.LC3/TUNEL double-positive cells accounted for about34.3%of LC3 plus TUNEL-positive cells. This study suggests that the protective effects of GDNF and HGF were greatly associated with not only the antiapoptotic but also the antiautophagic effects;maybe two types of cell death can occur in the same cell at the same time, and GDNF and HGF are capable of ameliorating these two pathways.V C2010Wiley-Liss,Inc.Key words:apoptosis;autophagy;GDNF;HGF; cerebral ischemiaIn ischemic diseases, ischemic severity and duration lead to cell death and determine tissue pathology(Cotran et al.,1998).In recent years, new types of cell death have been described;three types of cell deaths have been distinguished mainly by morphological criteria.Type I cell death is better known as apoptosis, and autophagic vacuoles inside the dying cell are typical for type II cell death, whereas type III cell death(better known as ne-crosis)is distinguished by early plasma membrane rup-ture and dilation of cytoplasmic organelles(Kroemer, 2005;Galluzzi,2007;Feig and Peter,2007).Apoptosis is associated with nuclear and chromatin condensation, DNA fragmentation, organelle swelling, cytoplasmic vacuolization, and nuclear envelope disruption(Kerr et al.,1972;Green and Kroemer,1998).Autophagy is a regulated process of degradation and recycling of cellularContract grant sponsor:Ministry of Education,Science,Culture and Sports of Japan;Contract grant number:21390267;Contract grant spon-sor:Reasearch Committee of CNS Degenerative Diseases(to I.Nakano); Reggiori and Klionsky,2002;Klionsky et al.,2003; Yoshimori,2004).Recently,27autophagy-related (ATG)genes were identified whose products appear to be related to the autophagy process. These genes were characterized in yeast(Klionsky et al.,2003;Yorimitsu and Klionsky,2005).For example, rat microtubule-asso-ciated protein1light chain3(LC3),a mammalian homologue of Atg8,plays a critical role in the formation of autophagosomes(Kirisako et al.,1999).Glial cell line-derived neurotrophic factor(GDNF), a member of the transforming growth factor-b super-family(Lin et al.,1993),has a potent neuroprotective effect on a variety of neuronal damage both in vitro and in vivo(Lin et al.,1993,1995;Beck et al.,1995;Tomac et al.,1995;Henderson et al.,1997).We and others have reported that topical application and intracerebral administration of GDNF decreased the size of ischemia-induced brain infarction and the number of TUNEL-positive neurons with suppressing apoptotic pathways such as caspases-1and-3(Abe et al.,1997;Wang et al., 1997;Kitagawa et al.,1998).Hepatocyte growth factor(HGF)is a multifunc-tional growth factor originally identified as a potent mitogen for hepatocytes in primary culture(Nakamura et al.,1984;Russell et al.,1984).HGF specifically binds and activates a tyrosine kinase receptor encoded by the c-Met protooncogene(Bottaro et al.,1991).Both HGF and c-Met were reported to be expressed in both adult and fetal central nervous system(Matsumoto and Naka-mura,1997;Maina and Klein,1999).HGF plays impor-tant roles in mitogenesis, motogenesis, morphogenesis, antiapoptosis, and angiogenesis(Nakamura et al.,1989; Zarnegar and Michalopoulos,1995;Matsumoto and Nakamura,1996;Maina et al.,1998;Van Belle et al., 1998).In the brain,HGF has the functions of a neurotro-phic factor via promotion of antiapoptosis, neurite exten-tion,and migration;a modulatory factor for glial cell numbers and function;and an angiogenic factor(Honda et al.,1995;Van Belle et al.,1998;Miyazawa et al., 1998;Sun et al.,2002).We and others have reported that intraventricular and intracerebral administration of HGF protein or transplantation of bone marrow stromal cells with ex vivo HGF gene transfer decreased the size of is-chemia-induced brain infarction(Miyazawa et al.,1998; Zhao et al.,2006)and the number of TUNEL-positive neurons with increasing presentation of Bcl-2(Tsuzuki et al.,2001).In addition,HGF also plays a role in attenu-ating ischemia-induced brain infarction via caspase-inde-pent cascade by preventing apoptosis-inducing factor (AIF)translocation downstream of poly(ADP-ribose)po-lymerase(PARP)and p53(Niimura et al.,2006a,b).However,the antiautophagic effect of GDNF and HGF in ameliorating ischemic brain injury has not yet been documented. In this study,we examined the antia-poptotic and antiautophagic effects of GDNF and HGF after tMCAO in rats.MATERIALS AND METHODSSurgical PreparationAdult male Wistar rats(SLC,Shizuoka,Japan)weighing 250–280g were used for the experiments. The animals wereanesthetized with an intraperitoneal injection of40mg/kgpentobarbital and positioned in a stereotaxic operating appara-tus.A2-mm-diameter burr hole was carefully made at3mm dorsal and4mm lateral to the right from bregma using anelectric dental drill, avoiding traumatic brain injury.Duramater was preserved at that time. The location of the burr hole was in the upper part of the right middle cerebral artery(MCA)territory. Then, the incision was closed, and the ani-mals were allowed to free access to water and food at room temperature. On the next day,at about24hr after the drilling, therats were lightly anesthetized by inhalation of a69%/30%(v/v)mixture of nitrous oxide/oxygen and1%halothane using a face mask.A midline neck incision was made and the rightcommon carotid artery exposed, and then inhalation of anes-thetics was stopped. When the animal began to regain con-sciousness,the right MCA was occluded by insertion of4-0surgical nylon thread with silicone coating through the com-mon carotid artery(Nagasawa and Kogure,1989;Abe et al., 1992).With this technique, the tip of the thread occludes theorigin of the right MCA.The reliability of producing success-ful strokes is almost complete in this model(Koizumi et al., 1986).During these procedures, body temperature was moni-tored with a rectal probe and maintained at37860.38Cusing a heating pad.The surgical incision was then closed, and the animals were allowed to recover at room temperature. After90min of MCA occlusion(MCAO),cerebral bloodflow(CBF)was restored by removal of the nylon thread. HGF or GDNF Treatment In VivoJust after restoration of CBF,the dura mater under the burr hole was carefully removed, and a small piece(8mm3) of spongel(Yamanouchi Pharma Co.,Ltd.)presoaked in9l lRinger solution(Otsuka Pharma.Co.,Ltd.)as vehicle or a solution containing GDNF(3.0l g in9l l of vehicle;Sigma,St.Louis,MO)or HGF(30.0l g in9l l of vehicle;KringlePharma)was placed in contact with the surface of the cerebralcortex. In terms of the dose of HGF,5.0,10.0,and30.0l g were checked in our preliminary experiment(Miyazawa et al.,1998;Niimura et al.,2006a,b),and the effect of30.0l g in9 l l of vehicle was the best among them. The dose of GDNF was chosen based on our previous report(Abe et al.,1997).The spongel was buried in the skull bone. The surface of theskull bone was then covered with vinyl tape, and the head skin incision was closed. It is reported that application of growth factor in spongel is as effective as chronic infusion (Otto et al.,1989).The above-described operations were per-formed in a sterile fashion.Sham-operated control animals underwent burr hole surgery, exposure of the common ca-rotid artery without MCAO,and placement of spongel pre-soaked in vehicle. The experimental protocol and procedures were approved by the Animal Committee of Okayama Uni-versity School of Medicine.2198Shang et al.Journal of Neuroscience ResearchPreparation and Quantitative Analysis ofInfarct VolumeThe animals(n55)were sacrificed at24hr after the restoration of CBF under deep anesthesia with pentobarbital (10mg/250g rat). The rats were transcardially perfused with heparinized saline, followed by4%paraformaldehyde in phos-phate buffer(PB).The whole brain was subsequently removed and immersed in the samefixation for12hr at48C.After washing out of paraformaldehyde by PB,the brain was immersed in sucrose solution in PB and then rapidly frozen in powdered dry ice and stored at–808C.Coronal brain sections of20l m thickness were prepared by using a cryostat and mounted on a silane-coated glass. For quantitative analysis of infarct volume, the sections were stained with hematoxylin and eosin(HE)and observed with a light microscope(Olympus BX-51;Olympus Optical). The area of the infarct was measured infiver sections by pixel counting using a computer program for Photoshop7.0,and the volume was calculated.Western Blot AnalysisWestern blot analysis was performed using the infarct hemisphere offive mice from each group. We added3ml cold lysis buffer(50mM Tris-HCl,pH7.2,10%glycerol, 250mM NaCl,0.1%NP-40,2mM EDTA,and protease inhibitors)to the brain tissue and homogenized it at48C.The homogenate was centrifuged at12,000rpm at48C,and the supernatant was used for Western blotting. We carried out Western blot analysis using standard techniques with an ECL Plus detection kit(GE Healthcare). The dilution of the anti-LC3(Medical Biological Laboratories;No.PD012)antibody was1:500.We carried out densitometry analysis in Scion Image Beta4.02software and took the average of thefive mice.TUNEL StainingIn accordance with our previous report(Abe et al., 1997),TUNEL study was performed using a kit(Roche, Nonnenwald,Germany)that detects double-strand breaks in genomic DNA with diaminobenzidine.Single Immunofluorescence AnalysisThe fresh-frozen sections werefixed for10min in ice-cold acetone and air dried. Then, the sections were rinsed three times in PBS(pH7.4).After blocking with10%normal rabbit serum for2hr,the slides were incubated for16hr at 48C with thefirst antibody:anti-LC3antibody(MBL;No. PM046)at1:200or anti-p-mTOR antibody(Cell Signaling Technologies,Danvers,MA;No.2971)at1:100diluted in PBS containing10%normal rabbit serum and0.3%Triton X-100.To confirm the specificity of the primary antibody,a set of sections was stained in a similar way, without primary anti-bodies.The sections were then washed and incubated for2hr with the second antibody:Texas red-labeled anti-rabbit IgG antibody at1:200(Vector,Burlingame,CA)or FITC-labeled anti-rabbit IgG antibody at1:200(Vector).The slides were then covered with Vectashield Mounting Medium with40,60-diamidino-2-phenylindole(Vector).Double-Immunofluorescence AnalysisDouble-immunofluorescence studies were performed for LC3plus neuronal nuclear antigen(NeuN)or glialfibrillary acidic protein(GFAP)or TUNEL or N-acetylglucosamine oligomers(NAGO).Lycopersicon esculentum lectin(LEL)is a glycoprotein with affinity for NAGO,which mature vascu-lar endothelial cells express(Augustin et al.,1995).The stain-ing steps were the same as those described above;LEL and thefirst antibody were chosen with each dilution as follows: anti-LC3antibody at1:200,anti-NeuN antibody(Chemicon, Temecula,CA)at1:200,anti-GFAP antibody(Dako,Carpin-teria,CA)at1:1,000,biotinylated LEL(Vector)at1:200 diluted in PBS containing10%normal rabbit serum and0.3% Triton X-100.The second antibody were Texas red-labeled anti-rabbit IgG antibody at1:200(Vector)plus TUNEL enzyme and label or FITC avidin D(Vector)or FITC-labeled secondary antibodies(Vector). The treated sections were scanned with a confocal microscope equipped with an argon and HeNe1laser(LSM-510;Zeiss,Jena,Germany).Sets of fluorescent images were acquired sequentially for the red and green channels to prevent crossover of signals from green to red or from red to green channels.Quantitative AnalysisTo evaluate the results of TUNEL staining and single-immunofluorescence analysis quantitatively, the positively stained cells were counted in the cerebral cortex at the boundary zone infive coronal sections per rat brain. In the double-fluorescence studies, the double-positive cells and ves-sels were counted in the same manner. Results are expressed as means6SD.Statistical AnalysisAll data are expressed as means6SD.One-way ANOVA with post hoc test was used for each evaluation.RESULTSQuantitative Analysis of Infarct VolumeNinety minutes of MCAO caused large infarcts of the lateral cortex and the underlying caudoputamen. Infarct areas offive coronal sections(2,4,6,8,and10 mm caudal from frontal pole)are shown(Fig.1a).The infarct volume of the vehicle-treated group was478.06 34.8mm3(mean6SD),HGF-treated group was420.0 623.8mm3,and GDNF-treated group was306.06 52.2mm3.The infarct volume of GDNF(**P<0.01)-and HGF(*P<0.05)-treated groups was significantly smaller than that of vehicle-treated group(Fig.1b).Antiapoptotic EffectThe TUNEL-positive stained cells were distributed in the cerebral cortex and dorsal caudate of the MCA territory but were not found in other areas of the ipsilat-eral hemisphere or the contralateral side. The staining was found exclusively in the nuclei of neuronal cells.A strong staining for TUNEL was present in the vehicle-treated group, but the treatment with GDNF(**P< GDNF and HGF Effects in tMCAO Rats2199Journal of Neuroscience Research0.01)and HGF (*P <0.05)greatly reduced the number of TUNEL-positive cells at 24hr after 90min of MCAO:389.8663.7/mm 2in vehicle-treated group,247.5654.5/mm 2in HGF-treated group,76.2632.1/mm 2in GDNF-treated group (mean 6SD;Fig.2a,b).Antiautophagic EffectExamining the intracellular localization of LC3in the ischemic brain.Cerebral ischemia indu-ces autophagy (Adhami et al.,2006;Rami et al.,2008).To show whether the protective effects of GDNF and HGF are associated with the antiautophagic effect after cerebral ischemia,we examined the protein level of LC3in ischemic and normal brains (Fig.3a).The number of cells displaying punctate LC3fluorescence significantlyincreased after cerebral ischemia, and the receptors consisting of the Ret receptor tyrosine kinase plus a glycosylphosphatidylinositol-linked core-ceptor termed GDNF family receptor a 1(GFR a 1).After binding to its specific receptor complex,GDNF activates several downstream intracellular pathways, including phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt;Soler et al.,1999)and extracellular signal-regulated kinase 1/2/mitogen-activated protein kinase (ERK1/2MAPK)pathways (Worby et al.,1996).Signals through PI3K/Akt or ERK1/2MAPK pathways lead todifferentFig.4.Western blot analysis of infarct hemisphere for al.,1999;Vaillant et al.,1999),whereas the ERK1/2MAPK pathway is involved in neuronal differentiation(Perron et al.,1999).HGF activates the protooncogenic receptor tyrosine kinase c-MET and subsequent downstream pathways, including the antia-poptotic protein kinase-B(PKB/Akt)cascade,the prolif-erative MAP-kinase pathway(ERK1/2and p38MAPK), and the STAT3signaling(signal transducers and activa-tors of transcription;Birchmeier et al.,2003;Okano et al.,2003;Hanada et al.,2004).Similarly,activation of the PI3kinase/Akt pathway, which is a well known way to inhibit apoptosis, also inhibits autophagy(Arico et al.,2001).Thus,perhaps GDNF and HGF signifi-cantly reduced infarct size associated with both the antia-poptotic and the antiautophagic effects(Fig.1).From the results of TUNEL staining(Fig.2), GDNF and HGF had significant antiapoptotic effects. Autophagy wasfirst described in the1960s(Stromhaug and Klionsky,2001;Kundu and Thompson,2008),but many questions about the actual processes and mecha-nisms involved remain to be elucidated. In multicellular organisms, autophagy is a homeostatic mechanism that may represent thefirst stage in a cellular/tissue response that can range from autophagy to apoptosis and necrosis (Klionsky and Emr,2000).In this sense, autophagy is a protective mechanism that can eliminate cells that would otherwise prove harmful to the organism. In fact, autophagy has been suggested to play a critical role in type II(nonapoptotic)programmed cell death(Klionsky and Emr,2000).To show whether the protective effects of GDNF and HGF are associated with the antiautopha-gic effect after cerebral ischemia,we examined the pro-tein level of LC3in ischemic and normal brains.LC3is associated with autophagosome membranes after post-translational modifications.A C-terminal fragment of LC3is cleaved immediately after synthesis to yield a cy-tosolic form called LC3-I(18kDa).A subpopulation of LC3-I is further converted to an autophagosome-associ-ating form,LC3-II(16kDa).It was reported that the amount of LC3-II was correlated with the extent of autophagosome formation(Kabeya et al.,2000,2004). The conversion of LC3-I into LC3-II is accepted as a simple method for monitoring autophagy(Mizushima, 2004;Klionsky et al.,2007).We probed brain sections with an LC3antibody that detects both forms of LC3, and strong LC3-immunopositive puncta were observed in ischemic brains(Fig.3a,b).LC3can under some con-ditions be incorporated into protein aggregates(Kuma et al.,2007).This is in accordance with Rami et al. (2008),and we found that LC3is expressed mainly in neurons in the cerebral cortex and dorsal caudate of the MCA territory(Fig.3c,d).Western blot analysis of LC3was performed in our study as well, and we detected that the amount of LC3-I plus LC3-II was significantly higher after cerebral is-chemia and that the amount of LC3-II change was cru-cial in that(Fig.4).The number of LC3-positive cells or the amount of LC3was significantly decreased with GDNF and HGF treatments.Mammalian target of rapamycin(mTOR)is a phosphatidyl inositol kinase-related kinase that negatively regulates autophagy(Schmelzle and Hall,2000;Yori-mitsu and Klionsky,2005).It has been reported that mTOR function is activated by phosphorylation of Ser2448(Nave et al.,1999;Ravikumar et al.,2003). Recent studies have presented evidence that insulin sig-nal stimulates phosphorylation and activity of mTOR via Akt/PKB signaling(Schmelzle and Hall,2000).In our study,we confirmed that the level of p-mTOR,an activated form, increased in the HGF and GDNF treated groups(Fig.5).It can also be hypothesized that GDNF and HGF partially regulated autophagy by the mTOR intracellular signaling pathway. The results described above suggested that both GDNF and HGF have signifi-cant antiautophagic effects. Our study suggests that the protective effects of GDNF and HGF were greatly asso-ciated both with antiapoptotic and with antiautophagic effects in terms of reducing the number of TUNEL-pos-itive cells,LC3,and activating mTOR.The number of LC3/TUNEL double-positive cells was about34.3%of LC3plus TUNEL-positive cells after cerebral ischemia (Fig.6),which suggested that perhaps one cell can undergo apoptotic cell death or autophagic cell death or two types of cell death at the same time. Thus,it is pos-sible that there are relations between apoptosis and autophagy. It was reported that apoptosis regulator also interacts physically with an autophagy regulator;for example,Beclin1can prevent autophagy induction. Beclin1was identified as a Bcl-2interacting protein (Liang et al.,1999),and Beclin1also interacts with the other major antiapoptotic Bcl family protein(Bcl-xL) and increases autophagy in nutrient-deprived or growth-factor-withdrawn cells, allowing cell survival(Boya et al.,2005;Lum et al.,2005)by inhibiting apoptosis. In this regard, perhaps in the future GDNF and HGF will be used in cerebral ischemia to protect against neuronal damage. We propose that both GDNF and HGF could provide important therapeutic benefits in terms of not only antiapoptotic but also antiautophagic effects for acute stroke patients. However, when gelfoam is chosen for the delivery of these factors, the amounts of GDNF and HGF should be carefully redetermined before clini-cal use of these proteins, because better neuroprotective effects in cerebral ischemia have been proved when these proteins are applied intrastriatally or intraventricluarly using an osmotic pump(Miyazawa et al.,1998;Tsuzuki et al.,2000).Differential roles of GDNF and HGF in chronic cerebral ischemia are of great interests and are bcurrently under investigation in terms of the presence of various extraneurotrophic activities of HGF(Funa-koshi and Nakamura,2003).ACKNOWLEDGMENTSWe thank Kringle Pharma(Osaka,Japan)for the gift of HGF.The authors declare that they have no competingfinancial interests.2204Shang et al.Journal of Neuroscience Research。
