建立同时符合中医“心肾不交”和下丘脑-垂体-肾上腺轴紊乱的失眠大鼠模型。方法

采用母乳鼠分离

法、多种焦虑刺激和腹腔注射对氯苯丙氨酸造失眠模型,并考察受试大鼠的一般行为学、血象及血浆Orexin 、促肾上腺皮质激素释放激素(CRH )、促肾上腺皮质激素(ACTH )的水平。结果模型组大鼠睡眠时间显著减少,目光呆

滞,双目、口唇呈苍白色,爪甲较光滑,皮毛重度枯槁蓬乱;血象检查血浆中红细胞数目和血红蛋白水平显著降低;

血浆中ACTH 、Orexin 、CRH 水平显著性升高。结论本实验的失眠大鼠模型能体现了“心肾不交”所致下丘脑-垂

体-肾上腺轴内分泌紊乱的失眠发生特点。

Rat model for insomnia caused by heart-kidney imbalance

ABSTRACT :AIM To establish a rat model for insomnia that was consistent with both heart-kidney imbalance

and disorder of hypothalamic-pituitary-adrenal (HPA )axis.METHODS

Maternal separation ,multiple anxiety

stimulation ,and the abdominal administration of chlorophenylalamine were adopted to induce rats with insomnia.The general behavior of rats was observed and their hemogram and the levels of ACTH ,orexin ,CRH were meas-ured.RESULTS

Compared to the control group ,the rats in the model group showed sleep-less ,glassy-eyed ,

pale lips ,and withered fur.The number of red blood cells and the hemoglobin concentration in plasma decreased significantly as well ,while the level of ACTH ,orexin and CRH significantly increased.CONCLUSION The in-somnia rats can present the symptom of heart-kidney imbalance and dysfunction of hypothalamic-pituitary-adrenal

axis.
